Production and biochemical characterization of xylanases synthesized by the thermophilic fungus Rasamsonia emersonii S10 by solid-state cultivation

چکیده

The xylanolytic enzyme complex hydrolyzes xylan, and these enzymes have various industrial applications. goal of this work was to characterize the endoxylanases produced by thermophilic fungus Rasamsonia emersonii in solid-state cultivation. Tests were carried out evaluate effects pH, temperature, glycerol phenolic compounds on activity. Thermal denaturation one isolated evaluated. crude extract from R. applied breakdown pretreated sugarcane bagasse, quantifying release xylose glucose. optimum pH value for enzymatic 5.5, 80 �C temperature. Regarding stability extract, highest values occurred between ranges 4 5.5. Several tested, showing an increase activity except tannic acid. Zymography displayed four corresponding endoxylanase bands, which extraction a polyacrylamide gel. thermodynamic parameters Xylanase C evaluated, half-life greater than 6 h at (optimum temperature), addition high melting temperature (93.3 �C) structural resistance thermal denaturation. Pretreated bagasse has good hemicellulose conversion xylose.
